Specificity

Clone DCS-22 is a mouse monoclonal antibody that detects human Cyclin D3. It targets an epitope within 20 amino acids from the C-terminal region.

Target description

G1/S-specific cyclin-D3 (UniProt: P30281; also known as Cyclin D3) is encoded by the CCND3 gene (Gene ID: 896) in human. Cyclin D3 is one of the most widely and ubiquitously expressed D-type cyclin. It serves as a growth factor sensor and a regulatory component of the cyclin D3-CDK4 complex that phosphorylates and inhibits members of the retinoblastoma protein family, including RB1, and regulates the cell-cycle during G1/S transition. Phosphorylation of RB1 allows dissociation of the transcription factor E2F from the RB/E2F complex and the subsequent transcription of E2F target genes which are responsible for the progression through the G1 phase. Cyclin D-CDK4 complexes are major integrators of various mitogenic and antimitogenic signals. They accumulate at the nuclear membrane and are then translocated to the nucleus through interaction with KIP/CIP family members. Cyclin D3 is also shown to be essential for cell proliferation stimulated by TSH and TSH induces the nuclear translocation of D3- CDK4 complex. TGF-beta is reported to block TSH induced nuclear translocation of D3-CDK4 complex. Elevated levels of cyclin D3 have been reported in breast cancer and are indicator of poor survival.
